Cover: colocalization of endocytosed surfactant protein (SP) A and lipid and of early endosomal markers in type II cells by confocal laser scanning microscopy. Top left: type II cells were incubated with unlabeled SP-A and rhodamine-phosphatidylethanolamine-labeled liposomes (red) for 3 min at 37°C and then incubated at 3°C for 30 min with FITC-labeled concanavalin A to visualize the cell boundaries, demonstrating true internalization of the lipid. Top right: to demonstrate colocalization (yellow) of SP-A with clathrin, type II cells were incubated with FITC-labeled SP-A (green) for 3 min at 37°C and subsequently with anti-clathrin antibody and cyanine-3-labeled anti-mouse IgG (red). Bottom left: colocalization (yellow) of FITC-labeled SP-A (green) internalized for 3 min and an antibody against the early endosomal marker EEA1 (red).
