Cover: Activation of a mitogen-activated protein kinase extracellular signal-regulated kinase (ERK) 2 in response to activation of MUC1 as assessed by immunofluorescence staining. Stable CD8/MUC1 cells (top) and transiently transfected COS7 cells (bottom) were treated with (right) and without (left) anti-CD8 antibody followed by two-color immunofluorescence analysis. Dual staining of both the anti-CD8 (green) and the anti-ppERK1/2 (red) antibodies in the cytoplasm and nucleus indicates that ERK2 can be activated after treatment with the anti-CD8 antibody (right). However, transfected but nonactivated cells do not show ERK2 staining (left). Untransfected COS7 cells treated with the anti-CD8 antibody also reveal no ERK2 activation (stained blue with 4',6-diamidino-2-phenylindole for its nucleus; bottom right).
